TrichoCHIT: the application of DNA BarCode for screening of superior chitinase producers among custom set of H. lixii/T. harzianum strains
using Hypocrea lixii/Trichoderma harzianum and chitinase production in solid state fermentation (SSF) using wheat bran and crude chitin as a substrate, we tested, whether fungal strains with superior enzyme production may be diagnosed by DNA BarCodes. A statistically supported superior chitinase production was obtained for strains carrying one of the observed ITS1 and 2 and tef1 alleles corresponding to T. harzianum ex-type strain CBS 226.95.
TrichoCHIT is a DNA BarCode tool for rapid identification of these strains.
Note: ITS1 and 2 or tef1?
if your sample includes different species of Hypocrea/Trichoderma we recommend you to sequence ITS1 and 2 and use TrichOKEY for the species identification.
The superior chitinase producers will be identified by TrichOKEY as "Trichoderma harzianum sensu stricto".
If you screen the set of already identified H. lixii/T. harzianum strains for the potential superior chitinase producers we recommend you to sequence the 4th_large intron of tef1 and use the form below:
